Spencer was born in Melbourne, Australia. He is the son of Dr. Rodney and Robyn Spencer, the founders of the political party Australians Against Further Immigration. His mother was also a prominent supporter of Pauline Hanson's One Nation Party. Spencer has two brothers, Tarney and Luke, and a sister, Polly. He attended Canterbury Primary School until Grade 1, followed by Malvern Central School through year 8. He attended Scotch College from year 9 and received an audition for the show "Neighbours" during year 10. He continued to work toward VCE while working.

Initially better known in Australia and the UK as Billy Kennedy in the Australian soap opera Neighbours, a role he played from 1994 to 2000, he has since appeared in the drama Death in Holy Orders, as well as the movies, Uptown Girls and Swimming Upstream.
As of 2004. he is based in the USA playing the role of Dr. Robert Chase in House, alongside Hugh Laurie in the title role. Jesse also recently co-starred with his House co-star, Jennifer Morrison, in the movie Flourish.
Although many Australian actors have found success in American film and television, Spencer is noteworthy as being one of the very few to actually play an Australian character in an ongoing core role.

- Spencer has extensive family ties to the medical profession: his father Rodney, two brothers, Tarney and Luke, and sister, Polly are doctors in Australia. When watching House, Spencer's brothers try to diagnose the problem based on the symptoms and medical jargon presented in the show. They also look for medical slip-ups in the dialogue.
- Spencer also has links to doctors through his acting jobs. Spencer played character, Billy Kennedy, son of local Neighbours Doctor, Karl Kennedy which is noted on the official neighbours website as part of the various past storylines.
